The Spain international suffered a hamstring injury earlier in the month against Atletico Madrid and will not manage to return for the clash with Barcelona.
A scan on Wednesday morning revealed that Negredo will not be able to return to training with the rest of the squad before Friday.
The 26-year-old should be fit for the La Liga clash with Racing Santander later this month.
